Ad Hoc, if you don't have much time, and still want to see a lot of locations in the city, here's a nice walking tour I made up for you. All the locations are within walking distance, and the whole walk won't take up more than an hour and a half max.
I'd start with the Phoenix Building. It's very easy to find, in West Georgia Street, which is in downtown Vancouver. Just walk down the street toward Stanley Park (which has some other nice locations) and then you'll see the Phoenix Building from afar. Although several buildings have been built around it compared to when the series was made, it still stands out .
(The first time I was in Van I stayed with friends who lived right next to the Phoenix Building. Quite exciting.) From there it's no more than a two minute walk down to Coal Harbour where the houseboat used to be. The Marina hasn't changed very much, and you'll recognize several buildings there – like the apartment hotel that is often seen in the background in the series.
From the marina it is a very short walk to Trade & Convention Center at Canada Place. Just walk along the waterfront, keeping the buildings on your right side, and in a few minutes you'll walk right into the Convention Center, where scenes from episodes like 'Lost Love' and 'Runners' have been shot. Walk a bit farther and then on your right you'll see the famous Gas Clock, seen in the later intros of MacGyver episodes.
If you keep walking along the waterfront, you'll see another location not far from there: the Burrard Bridge, at the end of Burrard Street. Under the bridge scenes from 'There but for the grace' and 'Tough boys' have been shot.
Walking back into the city on Burrard St. you'll end up at the Hyatt Towers Hotel. Across from the hotel on Burrard is the church of which the exterior is seen in 'Legend of the Holy Rose.'
From there it's an easy walk to the heart of the city, where you'll find The Vancouver Art Gallery on Hornby Street. This building, and the fountain in front, have been used in several episodes, like 'the Battle of Tommy Giordano', 'The Negotiator' and 'Lesson in Evil'.
Across from the Art Gallery you'll see a big glass building with the huge pendulum inside, used in 'Fire and Ice'. That's the HSBC Bank. Now you're back on W.Georgia Street, and this is where the tour ends.
If you have the time to go a bit out of town, Steveston is the place I can highly recommend. In Moncton Street you'll find locations used in episodes like 'Jack in the box' and 'The Secret of Parker House'. And at the end of the street lies the Cannery used in 'Back from the Dead'.
If you don't want to go that far, you can take a skyride to Grouse Mountain (used in 'Cease Fire'). From Grouse Mountain you'll have a wonderful view over the city, and in the distance you can see the mountains that are called the The Twin Sisters, aka 'Los dos Pumas' in 'The Treasure of Manco'.
